益气解毒方对γ射线致小鼠小肠急性损伤的防护效应研究*

Protective effects of Yiqi Jiedu Decoction on γ-ray-induced small intestinal injury in mice*

摘要: 目的 探究益气解毒方预防给药对2.0 Gy 60Coγ射线致BALB/c小鼠小肠急性损伤的防护效应。 方法 成年期BALB/c雄性小鼠190只,按体重随机分组为正常组40只、模型组30只、阳性药对照组40只、益气解毒方高剂量组40只、益气解毒方低剂量组40只。正常组、模型组灌胃去离子水,阳性药及中药高、低剂量组分别灌胃安多霖悬浮液及益气解毒方高、低浓度药液,连续灌胃10 d。给药(水)10 d后,从正常组、阳性组、高剂量组、低剂量组中,照射前分别随机选取10只小鼠进行取材检测。然后除正常组外,其余小鼠接受2.0 Gy 60Coγ射线一次性全身照射,每组分别于照射后1、3、7 d随机选取10只小鼠取材。实验过程中每天记录小鼠体重变化,取材小肠组织分别进行HE病理切片染色、绒毛长度、隐窝深度测量,凋亡细胞TUNEL染色及计数,occludin mRNA表达检测。结果 分别预防给予小鼠高、低剂量益气解毒方后,小鼠体重增长良好;照射后,小鼠降低的体重能较快恢复。益气解毒方低剂量能升高照后小肠组织occludin mRNA的表达,减少射线导致的小肠细胞凋亡,促进小肠绒毛和小肠隐窝的恢复,从而减轻了射线对小肠组织造成的损害。结论 益气解毒方预防给药,对2.0 Gy 60Coγ射线致BALB/c小鼠小肠急性损伤具有良好的防护效应,其中低剂量防护效果较好。

Abstract: Objective To explore the protective effects of Yiqi Jiedu Decoction on small intestinal injuries caused by 2.0 Gy 60Co γ-ray in BALB/c mice. Methods 190 male BALB/c mice were randomly divided into the blank control group (n=40), irradiation group (n=30), positive medicine group (n=40), high-dose Yiqi Jiedu Decoction group (n=40) and low-dose Yiqi Jiedu Decoction group (n=40) . The NC and IR groups were given deionized water by gavage once daily, while positive medicine group, high-dose Yiqi Jiedu Decoction group and low-dose Yiqi Jiedu Decoction group were administrated with Anduolin suspension, high- and low-dose YiqiJiedu Decoction, respectively. After 10 days’pretreatment, 10 mice from each group (except IR group) were sacrificed respectively to test indicators. Then, the others (except NC group) were followed by whole-body exposure to 2.0 Gy 60Co γ ray. On the 1st, 3rd, 7th day of exposure, 10 mice from each group were sacrificed respectively and the body weight, pathological section and apoptosis of the small intestine, and the expression of occludin mRNA in the small intestine were measured. Results The body weight of mice reduced significantly after exposure, while both high-dose and low-dose Yiqi Jiedu Decoction promoted the recovery of body weight. Moreover, the small intestine structure was injured obviously after irradiation. The low-dose Yiqi Jiedu Decoction promoted the recovery of both intestine villi and intestine crypt. In addition, RT-PCR and TUNEL analysis showed that low-dose Yiqi Jiedu Decoction up-regulated the expression of occludin and reduced the apoptosis rate in the small intestine after irradiation. Conclusions Yiqi Jiedu Decoction has a good protective effect on acute intestinal injury caused by 2.0 Gy 60Co γ-ray in BALB/c mice, and low-dose Yiqi Jiedu Decoction has a better protective effect.
